J&J's Simponi (golimumab) is a fully-human IgG1 mAb specific for TNFa. It inhibits TNFa activity, thereby blocking the inflammatory activity of this cytokine and downstream reactions. Simponi is approved for patients with moderate to severe, active RA, and is typically prescribed in conjunction with MTX as a second line of therapy in patients who have failed treatment with another anti-TNF agent, or who have had an unsatisfactory response to MTX.
